網膜剥離後の細胞増殖におけるp27(KIP1)の役割

研究概要

これまで我々は野生型adultマウスにおいて、細胞周期関連蛋白の一つであるp27(KIP1)がMuller細胞および網膜色素上皮(RPE)細胞に発現していることを報告してきた(Yoshida et al.Anat Embryol 2004)。今回はマウス網膜剥離における細胞周期関連蛋白の発現と細胞増殖活性との関連について検討した。[材料と方法]生後12週のC57Bl6マウスを使用した。過去の報告を参照し網膜剥離を作製した(Yoshida et al.Graefes Arch Clin Exp Ophthalmol 2004)。剥離作製2日および4日後に還流固定し、眼球のパラフィン包埋標本を作製した。G1からS期への進行に関与するcyclinD1、細胞周期の進行停止に関与するp27(KIP1)について免疫組織化学的に検討した。細胞増殖活性の評価のためproliferating cell nuclear antigen(PCNA)の発現も検討した。[結果]正常ではp27(KIP1)はMuller細胞およびRPE細胞の核に発現されていたが、cyclinD1の発現はなかった。網膜剥離部のRPEでは剥離2日後からcyclinD1、PCNAの発現があり、一方p27(KIP1)の発現はなかった。Muller細胞ではp27(KIP1)の発現は2日後までみられたが、4日にはなかった。これとは対照的にPCNAは2日後で発現しておらず、4日後に発現していた。CyclinD1は2日から4日後にかけて発現していた。[結語]網膜剥離によりMuller細胞とRPE細胞は増殖活性を有し、これらはp27(KIP1)の分解、cyclinD1の発現を介する。
