研究概要

高親和性IgE受容体(FcεRI)は、アレルギー反応誘導の鍵となる役割を果たしている。FcεRIを構成するサブユニットのうち、β鎖は細胞内シグナルおよび受容体の細胞表面上への発現の両方を調節し、アレルギー反応を緻密に制御する分子である。
我々は既に、ヒトβ鎖遺伝子の第4イントロン中の転写制御エレメントに、転写因子MZF-1がFHL3を含む高分子量複合体を形成して結合し、β鎖遺伝子の発現を抑制することを報告した。また、GM・CSFによりMZF-1の発現のみならず、FHL3の核移行が誘導されることを見出した。
本研究においてはまず、MZF-1、FHL3を含む高分子量複合体の実体を明らかにするために、yeast three hybrid assayによりヒトcDNAライブラリーからこの複合体の構成因子を探索し、NFYを同定した。その結果、NFYがFHL3を介してMZF-1と相互作用し、三者複合体を形成することが明らかとなった。さらに、NFYおよびNFYと結合することが報告されているヒストン脱アセチル化酵素(HDAC)がβ鎖遺伝子の第4イントロン中のエレメントを含む領域と相互作用することをChIPアッセイにより示した。また、GM-CSF存在下で培養することにより、β鎖遺伝子と相互作用するピストンのアセチル化が低下することを明らかにした。一方、GM・CSF存在下で培養したヒトマスト細胞株において、実際にβ鎖の発現が低下することを定量RT・PCRにより確認した。以上の結果から、MZF-1、FHL3を介してNFYさらに且DACがβ鎖遺伝子にリクルートされ、HDACによりピストンが脱アセチル化されることによりβ鎖遺伝子の発現が抑制されることが示された。また、GM-CSF存在下では、MZF-1の発現とFHL3の核移行の誘導により且DACのβ鎖遺伝子へのリクルートが促進され、ヒストンの脱アセチル化が進行してβ鎖遺伝子の発現が低下することが明らかになった。
