研究実績の概要 |
Cucumber seedlings were used for heat shock [HS] treatment 50oC for 20 s and their response against powdery mildew fungi were observed. Similar to response of melon seedlings after heat shock treatment 50oC for 20 s against grey mold in previous report, cucumber seedlings were also protected against powdery mildew if inoculation was conducted 0- 24 hours after treatment. Reduction of disease index [%] during those inoculation period were 11.67; 35 and 50- 56.67; respectively in day observation of 4 – 6 compared to Mock [dipped in 25oC for 20 s]. Reduction of disease index was not shown when inoculation conducted 2 days after treatment. It is showed that plants responded in very early period toward heat shock treatment. Results of gene expression on cucumber after heat shock treatment showed that Peroxidase and HSP70 genes mainly increased after heat shock treatment and treatment followed by powdery mildew inoculation. This research showed that heat shock protein especially HSP70 contributed in the response of cucumber seedlings to be more resistant against pathogen infection of powdery mildew. As literatures wrote that when activated in stress pathway, heat shock transcription factors [HSFs] will be released from its binding to HSP genes and attach to HSE leading for transcriptional activation of defense related genes, therefore study on HSP inhibitors and HSF gene expression is being conducted.

理由
Both excellent and challenging points are found in this research process. The excellent points are, first, I found consistent data in inoculation test after heat shock treatment both in melon again grey mold [previous research] and cucumber against powdery mildew [current research]; second is that HSP70 played important role immediately after HS treatment. HSIR is very challenging as plant response very fast after treatment and it does not give long lasting effect, supposed that the mechanism works before or different from systemic acquired resistance.

今後の研究の推進方策 |
In near future, focus of the research will be on screening of concentration for HSP inhibitors. Currently, inoculation test was conducted in treatment of Geldanamycin 10; 5; and 1 µM; 17 AAG in 500; 200; 100 µM; compared to control [DMSO 1%] and heat shock treatment as reference. Observation of disease index is conducting now and up to 4 days after treatment the symptoms in all samples are still unseen.
