Research project proceeds according to the research plan with minor modification.
(1) Integration of the genomic and all transcriptomic data for Aurelia life stages and tissues (P1,S1,S2,S3,S4,J1,J2) has been completed.(2) Genome browser and associated transcriptomic databases were established and accessible to the researches and public. (3) Two articles were published in connection to the project. As we anticipated in the previous report the ChIPSeq approach (which nobody tried before in Aurelia) turned out to be difficult due to high concentration of DNAses in the somatic tissues of Aurelia (not a problem for the germline cells). Thus, instead of ChIPSeq we proceeded in FY2018 with the comparison of DNA methylation patterns between polyp and jellyfish stages which were originally planned for FY2019. As proposed in the original research plan we successfully used PacBio sequencer to measure kinetic variation during base incorporation (dependent on methylation status of the DNA). In addition to that we used NanoPore PromethIon sequencer for direct detection of DNA modification patterns. Data analysis is in progress and we plan to submit our new data for publication within FY2019. Despite of the initial difficulties with ChIPSeq we will proceed further with this approach as the data is important for comparison with Nematostella polyp satge.
