We have proposed a new method for DNA sequencing using solid-state nanopores. Prior to this research, no structural information of a DNA molecule had ever been read for solid-state nanopore resistive pulse sensing using an applied external electric field, mainly due to (i) fast DNA translocation speed, (ii) high temperature fluctuation and (iii) insufficient spatial resolution. These problems have been successfully resolved using our new method by replacing the electric field with a solute concentration gradient that provides diffusion current for isothermal DNA structure detection. We have obtained multi-levels of current blockage signals of dsDNA oligonucleotides indicating various nucleotide bases (first time in history). The results revive hopes for solid-state nanopore DNA sequencing.
