研究概要

1997年哺乳類時計遺伝子のクローニングにより生体リズム研究は新しい時代に入り、最初の10年間で発振の分子機構が解明された。その次の世代である本研究では、時計遺伝子の生体における生理的・病理的役割解明に注力した。時計中枢の視交叉上核では、G蛋白質シグナル伝達が細胞リズムの同期に関わる分子機構を解明した。また、時計遺伝子プロモーター結合の人工転写因子を創成し、概日リズムの位相変位に成功した。臓器の時計の研究では、肝臓代謝、膀胱排尿のリズム機構を分子レベル解明し、副腎ではアルドステロン生合成を担う時計に制御される新酵素を同定し、この異常が高血圧に至る機構を解明した。
