難治性慢性ウイルス感染症及び腫瘍に対する新規細胞治療システム

研究概要

ウイルス誘発疾患(HTLV-I/ATL、HIV I/エイズ、およびEBV/関連リンパ腫など)や非ウイルス性腫瘍に対しては、形態学的、遺伝学的、そして、分子生物学的手法を駆使し、原因ウイルスによる持続感染、リンパ増殖性/変性的な異常、および免疫反応の分子機構などを理解する必要がある。そして、薬理学的/免疫的治療を含む新規坑ウイルス戦略を開発を行う必要がある。このため私たちはまず、NOD/SCID/IL2Rγ/マウス(NOG)を開発し、無処理マウスで新しいATLの動物モデルを確立した。さらに、私たちはいくつかの新たに開発されたNF-kB抑制剤の効果を示した。NK細胞は抗原感作なしで、腫瘍細胞とウイルス感染細胞を認識して攻撃するが、私たちはNOGマウスと実際の患者で、肺がんや乳がんに対するNK細胞の効力と安全を立証した。次に完全なヒト型マウスの完成を目指して、ヒト臍帯血移植を行った。ヒトB細胞、T細胞、および他の単核細胞/大食細胞の誘導が見られ、造血幹細胞とリンパ濾胞様組織の発達も見られた。この系では、CCR5とCXCR4-HIV-1の顕著な感染が起こり、急激なウイルス血症に引き続いて120日後以上も遷延する持続感染が見られた。その上、高率のウイルス感染を示した動物ではHIV-1Env gp120とGag p24に対する両者の抗体を検出した。同様に、これらのヒト化マウスでは、EBV感染に非常に感受性が高いことも示し、それらがヒトでの持続性ウイルス感染の特徴についてかなり再現できることを示した。これらの結果は、難治性慢性ウイルス感染症や非ウイルス性悪性腫瘍に対する新規治療法、とくに細胞を用いた治療法の開発に有用であろう。
