G0期のp27安定性に関わる巨大複合体の解析

研究概要

G0期特異的に形成されるp27巨大複合体構成因子、及びその制御因子の同定・機能解析を目標として本研究を行った。p27はG0期から細胞周期に再進入する際にタンパク質分解により発現量が急速に減少する、重要な鍵分子である。このp27のSer10部位をAlaに置換した非リン酸化型ノックインマウスを作製・解析したところ、本来非常に安定であるG0期にp27タンパク質が不安定化し、p27発現量が低下することが明らかとなった。これはG0期に形成されるp27の巨大複合体が増殖シグナルであるPI3キナーゼ活性によって阻害される結果、p27が不安定化することが示唆された。本研究ではこのG0期特異的なp27の巨大複合体プロテオミクスを行い、その構成因子を明らかにするとこと、またこのG0期特異的な巨大複合体の阻害分子の同定を試みた。
まず293T細胞にFLAGタグを付加したp27を発現させ、PI3Kインヒビター処理により細胞周期を停止させた細胞、PI3キナーゼ活性化時それぞれに特異的に結合する分子群を、抗FLAG抗体カラムを用いて濃縮し、p27複合体プロテオミクスを行った。その結果、30種類弱のタンパク質をPI3Kインヒビター存在下のp27結合分子として同定することに成功したが、これらp27結合分子の多くは既知であり、残りの結合因子について解析した結果、これらの中にもG0期においてp27と複合体を形成する分子群を見出すことは出来なかった。しかしPI3キナーゼ活性化時特異的にp27とPP2A脱リン酸化酵素の結合することを発見した。PP2AはSer/Thr特異的脱リン酸化酵素であることから、G0期にはリン酸化Ser10部位がG0期特異的な巨大複合体を促進させるが、増殖刺激によりPI3キナーゼが活性化された状態ではPP2Aがp27のリン酸化Ser10を脱リン酸化し、巨大複合体の形成が阻害されてp27が分解されやすくなることが推測された。この仮説を検証するため、PP2Aの阻害剤やsiRNAによる阻害がp27の分解(G0期からの脱出)に影響を与えるか検討している。
