研究実績の概要 |
The research purpose is to synthesize new amphiphilic polymers for efficient mRNA transfection in immortal T cells (Jurkat cells). Precursor polymers, poly(beta-benzyl-L-aspartate) (PBLA) with a degree of polymerization (DP) 21 was prepared by a ring-opening polymerization.The previous results demonstrated that the cyclohexylethyl amine (CHE) and diethylenediamine (DET) moieties-bearing amphiphilic polymer induced high messenger RNA (mRNA) transfection efficacy in vitro. Accordingly, new aliphatic amines with ring structure were selected here as next candidate moieties. The new aliphatic amines and DET were simultaneously reacted with PBLA. The obtained results showed that the introduction ratio of the aliphatic amines was lower than expected, suggesting that the synthesis condition needs to be optimized in the next fiscal year.

今後の研究の推進方策 |
The synthesis of amphiphilic polymers was little bit slower than the initial schedule because the previous synthetic methods did not provide sufficient introduction rate of hydrophobic moiety. This is because some aliphatic amines did not have a sufficient purity, resulting in the low reactivity with the precursor polymer. The polymer synthesis will further be optimized to obtain proper hydrophobic introduction rates. The feeding ratio and reaction time will be further altered to increase the introduction raties of hydrophobic moiety.
