A total of 189 Atlanto-Mediterranean individuals of Savalia savaglia have been shipped at the host institution by February 2019 and DNA extractions were performed testing different kits and protocols. Two specimens will be used for Whole Genome Sequencing (WGS). Three unidentified specimens of Savalia collected off Okinawa have been also considered for genetic comparisons. A four-days field work in Kume Island was performed in order to collect as many colonies as possible of Umimayanthus spp. Observations in the field confirmed the preliminary findings, according to which U. champuru colonises encrusting sponges of different species. In February 2019, diving surveys were conducted in eastern Okinawa with the main goal to select the best site(s) for species diversity and abundance.

理由
Concerning S. savaglia, about 75% of the specimens have been until now processed. In particular, the DNA extraction of 140 samples out of 189 has been performed testing different kits and protocols. So far the extractions succeeded for 36 samples out of 140. Two samples with high DNA quality and yield have been designated for WGS. Regarding Umimayanthus, detailed information on the sampling sites as well as the sampling strategy and samples fixation have been carefully planned. Unfortunately, the sampling of Umimayanthus spp. has been so far hampered by the bad weather and sea conditions. However, the majority of the specimens needed for the molecular analyses will be collected by the end of Summer 2019 from two sites in eastern Oikinawa.

今後の研究の推進方策 |
DNA extraction of the remaining S. savaglia individuals will be performed by the end of May. DNA extractions will be eventually repeated in order to obtain high DNA yield and quality from at least ten individuals each population. Three different molecular markers (COI, 16S rRNA and ITSs) will be amplified and sequenced for few individuals of different Mediterranean and Eastern Atlantic populations. Ninety-five (95) S. savaglia individuals will be used for library preparation and Restriction site Associated DNA Sequencing. The raw DNA data will used for SNPs calling. The obtained WG of the different S. svaglia individuals will be assembled and annotated testing multiple software. Regarding Umimayanthus, it will be explored the diversity of the host organisms (Porifera).
