Development of new theranostic nanoparticles, based on quantum dots (QDs), conjugated with multispin nitroxide-cyclodextrins for quantitative imaging assays of oxidative stress and redox-status of living cells and tissues. These nanoparticles will combine several features: 1) three imaging modalities (MRI, EPR, optical); 2) intracellular delivery and localization mainly into the mitochondria; 3) possibility to sense and visualize the areas of oxidative stress and redox-status in living cells and tissues; due to dynamics of their contrast properties, based on the balance between oxidizers and reducers; 4) redox-modulating activity and possibility to induce apoptosis in cancer cells, without significant effect on viability of normal cells; due to SOD-mimetic feature of nitroxide residue. 1. I synthesized and characterized of two quantum sensors using DLS, fluorescence spectroscopy, EPR spectroscopy, MRI on phantoms. I evaluated redox-sensitivity in model chemical systems. 2. I prepared MRI preclinical research environment. 3. I prepared cell and chemical research environment.
