研究実績の概要

It was confirmed that culturing pancreatic spheroids in a PDMS-chip leads to increase in ROS production compared with spheroids cultured in a plastic chip or in monolayer. Therefore, conditions for culturing pancreatic spheroids in PDMS-chip were optimised by adding antioxidant ascorbic acid-2-phosphate and comparing its action with two other antioxidants (n-acetylcysteine and dithiothreitol). The addition of antioxidants led to the increase in insulin production.
The possibility of formation of vascularised pancreatic spheroids in collagen gel was shown by embedding MIN6 and fibroblasts co-culture spheroids into collagen gel with dispersed pancreatic microvascular endothelial cells.

理由

As it was planned, I finished the work with antioxidants. The results were published in a paper and presented at international conference.
I also started the work with vascularised spheroids and neuro-vascularture.

今後の研究の推進方策

First, I want to investigate the influence of glucose concentration on MIN6 spheroids during their culture, as pancreatic beta-cells are known to be sensitive to glucose concentration.
Another direction is to continue investigate interactions between pancreatic, vascular and neuron cells. As my research showed, it is difficult to use PC12 (neuron cell line) for this purpose. Thus, I will need to learn how to work with primary or stem cells.
