In the frame of this project, we developed two new molecular sensors for: (i) Quantitative OxiStress In Vitro Assay & In Vivo Mapping; (ii) and Quantitative TRC In Vitro Assay & In Vivo Mapping; using MRI, EPR and fluorescence imaging. The probes are based on originally developed quantum dots (QDs, coated with multi-nitroxide- and triphenylphosphonium-conjugated cyclodextrins. Both assays are based on highly effective linear quenching of QD fluorescence by nitroxide radical, but not by its hydroxylamine form. This leads to dynamics of fluorescent/paramagnetic contrast, depending on the level of oxidative stress and redox-status of biological object. Both sensors were applied in vitro on cultured cells, characterized by different proliferating activity and respectively by different level of oxidative stress and redox capacity. TRC sensor was applied successfully in vivo for detection of redox status and oxidative stress in kidneys of mice – healthy and with hypercholesterolemia-induced renal dysfunction and inflammation. The final data were published recently in Analytical Chemistry (January, 2021) and press-released in Japan.
