研究実績の概要

In this study, we aimed to develop a multiple stem cell based therapy for stroke. To pursue such research goal our achievement so far are:
1)Development and characterization of microglia, neural and mesenchymal stem cell line.
2)Establishment of cell culture condition. Neural stem cells show some phenotypic differences depending on culture condition. Hence, several culture conditions were established to see which condition is good for stroke therapy.
3)To understand the mechanism, the interaction of transplanted cell types with each other, and with the host cells are important. Interaction with the host cells can be checked by transplanting them into the model animals. To investigate the interaction of transplanting cell types with each other, we established a double cell culture condition.

理由

Since we have established several cell lines and also established culture condition, currently we are conducting research on the interaction of stem cells in vitro condition:
1.Interaction of Neural stem cells and microglia in different culture condition regarding neuronal growth, survival and differentiation.
2.Interaction of neural stem cells and mesenchymal stem cells in different culture condition regarding neuronal growth survival and differentiation.
We are also conducting research on the in vivo interaction of those cells after transplantation with host brain cells in stroke animal models.

今後の研究の推進方策

After understanding the interaction of transplanted neural stem cells, microglia and mesenchymal stem cells, we have a plan to develop an effective cell-based therapy for stroke condition where neuroinflammation, restoration and regeneration of damaged neural tissue will be addressed. For this purpose, following matter should be taken into account:
1.Timing of transplantation- what is the best time for transplantation of a cell type
2.Sequence of cell transplantation- which cells should be transplantation first that will provide best outcome
3.Is there any harmful effect of these stem cells based transplantation- Since we are planning to transplant the cells intravenously, these cells may go to other organs besides brain. Then the interaction of these transplanted cell types in those organs should also be taken into account.

次年度使用額が生じた理由

Last year we had a plan to establish a suitable culture for neural stem cells, microglia and mesenchymal stem cells, and check their interaction in culture condition. Also we planned to transplant mesenchymal stem cells (MSC) and microglia in rat stroke model. However, to find a suitable culture condition took more time than expected. Consequently, we could not transplant the cells in animal stroke model. As a result, we could not spend the money allocated for animal model preparation and experimentation in last fiscal year.
Plan to use the incurring amount in this fiscal year: 1.Preparation of rat stroke model and transplantation of MSC and microglia 2.Check the functional neurological performances.
3.Check the pathological changes in stroke model after mesenchymal stem cell and microglia transplantation. 4.Check the molecular and cellular changes in stroke model after mesenchymal stem cell and microglia transplantation.
