In this study, we aimed to develop an improved therapy for stroke using stem cell transplantation. We successfully developed a stroke animal model and developed a stem cell therapy useful for the treatment of stroke. Here, first we transplanted IL-4 primed microglia, followed by neural stem cells (NSCs) intravenously in the stroke animal model. In previous years we found that IL-4 primed microglia and NSC co-transplantation animals have decreased tissue damage and cell infiltration, and increased NSC accumulation in stroke area. M2 type microglia in these co-transplanted animals were increased. Importantly, neuron and vessel numbers were increased in these co-transplanted animals. This year, we found that the cause of decreased tissue damage in co-transplanted stroke animals was due to phagocytic cleanup of the stroke area by microglia, many of them were transplanted cells. Also, single transplantation of IL-4 primed microglia decreased the damage of stroke area by phagocytosis of debris. Depending on these results, we are preparing a manuscript for publication in a scientific journal. Also, this year we published a paper where we showed the characteristics of the NSCs that we generated and used in this study.
