肝虚血再灌流障害に対する薬理学的ポストコンディショニング法の開発と分子機序の解明

研究概要

ラット肝虚血再灌流モデルで、PDEIII阻害薬、Rhoキナーゼ阻害薬の再灌流時投与によって保護効果を組織学的障害の有無とTUNELアッセイにより検討し、PDEIII阻害薬、Rhoキナーゼ阻害薬が薬理学的ポストコン作用を有することが明らかになった。PI3K-Akt活性化阻害薬wortmannin、LY294002とERK活性化阻害薬PD-98059、eNOS阻害薬L-NAMEを用いた検討ならびに免疫組織染色、ウエスタンブロット法によって、PDEIII阻害薬、Rhoキナーゼ阻害薬の薬理学的ポストコン作用に虚血ポストコンと同様にPI3-Akt、ERK、eNOSが関与していることが明らかとなった。
