In the past one year, we have made interesting progress regarding ARFC, a putative downstream effector for SB. As a transcription factor, we suspect that ARFC may regulate a range of transcriptional targets to mediate cell wall remodelling. To identify these targets, we performed RNA sequencing using an ARFC inducible line and pinpoint a range of cell wall related genes such as pectin methylesterases and expansins. We found that these genes are repressed by ARFC through Gene Ontology analyses. At the same time, through collaboration with Prof. Kotake (Saitama University) we have also confirmed that the cell wall compositions of ARFC knock-out mutants and ARFC overexpressor have changed, establish a link between ARFC and cell wall remodelling. In order to understand the expression pattern of ARFC, we have generated a promoter fusion reporter for ARFC. Using confocal imaging, we determined that ARFC expression is excluded from the stem cell. In our effort to prepare the sequencing population for our repressor mutants, Prince Charming (PC), we have established the somatic hybridization protocol in the lab. Results from my work has been presented at the Annual meeting of the Japanese Society of Plant Physiologists.
