研究実績の概要 |
We conducted many rats experiments. We bought the pre-diabetes rat model when compared with a normoglycemic population, which increases the risk of hippocampal memory dysfunction and type 2 diabetes mellitus (T2DM). We investigated whether the progression of spatial memory dysfunction is delayed with four months of exercise intervention since the pre-diabetic stage in an established model of T2DM rat. Memory function, biochemical, and molecular profiles were assessed in the pre-diabetic stage of OLETF rats, a model of T2DM, with LETO rats as genetic control. Then these rats were subjected to mild- or moderate-intensity running training with a treadmill for four months with repetition of same experiments. Significant dysfunction in hippocampal spatial memory was observed in the pre-diabetic stage of OLETF rats, accompanied by downregulated hippocampal MCT2 levels without alteration in hippocampal glycogen level. Four months mild or moderate exercise from pre-diabetes was effective in significant normalization of glycemic parameters with hippocampal molecular normalization through MCT2, glycogen, and BDNF levels. Finally, we observed the considerable improvement of memory dysfunction in early-T2DM rats. A four months exercise training regimen from pre-diabetes either in light- or moderate-intensity can delay the progression of spatial memory dysfunction with hippocampal MCT2-related-lactate-transport improvement of early-T2DM, implying a potential impact of mild exercise on T2DM. This is a preliminary finding, and we are continuing the experiments.

理由
The current research is on the right track both from the contexts of the timeline and the number of research activities scheduled to be done (100% done with target milestone achievement) with the proper management of project budget and funding. Keeping consistency with original research application and planning, in the first year of this project, we already performed many experiments on the pre-diabetes model. We confirmed that impairment of spatial memory dysfunction already started from the early pre-diabetes phase. Here we use the pre-diabetes phase of OLETF rats, which is an established model of type 2 diabetes. We tried to define the exercise intensity, especially for the mild exercise. Furthermore, we are successful in running the pre-diabetic rats with mild exercise intensity for four months and also able to demonstrate that this mild exercise regimen is beneficial to reverse the memory impairment and diabetes-related peripheral symptoms at early diabetes. Next, we plan to use this exercise regimen for other diabetic models and is possible for human subjects, and we did not face any difficulty in accomplishing the first-year plan for this research.

今後の研究の推進方策 |
In the 2nd year of this project, we would like to make much progress in a human study that we already started preparation at the end of the first year. This year, we will conduct several mobile health screening camps in rural Bangladesh and make an effort to diagnose diabetes who were no known cases of diabetes. We will screen diabetes by OGTT, in Bangladesh, in a rural setting, it is challenging to measure HBA1c for diabetes screening. And then, we conduct six months mild exercise program in these newly diagnosed diabetes cases and then check the improvement status on peripheral diabetes symptoms. Further, we will focus on VEGF angiogenic signaling cascade and the related apoptosis signaling cascade in the pre-diabetes hippocampus with or without exercise running. Besides, we tried to explore organ dysfunctions in other vital organs in early diabetes and the related effects of exercise.

年度の最終盤に、新型コロナウイルスの影響があり、ヒトの抗体検査用試薬キットを購入しなかったため。状況が落ち着き次第、試薬キットを購入する予定である。
