研究実績の概要 |
As for this year, we have planned to optimize the protocol for fabrication of the self-organized tissue ring. Our project progressed smoothly. The achievements are as following: 1. The self-organized tissue rings could be produced robustly. The traveling waves could be induced in more than 90% of all the rings. 2. The dynamic culture system has been established to promote the nutrient exchange in the tissue ring. The conduction speed of the action potential has been improved dramatically. 3. The contraction force, field potential, gene expression and electron microscope images have been obtained from the trained tissue ring to prove that the higher maturation of the cardiomyocytes. 4. One paper has been published on this project in this year.

理由
So far, we have almost achieved all the planned step for first year. Although there could be impact of the Coronavirus on our following experiments, we will try to cope with the negative impact and to make the project progress smoothly in the following year.

今後の研究の推進方策 |
Based on the previous work, we will carry out the following procedures regarding the transplantation application: 1.To further optimize the culture system to produce the tissue ring suitable for transplantation on animal heart model. To try to make the tissue ring into a sheet for better attachment on heart. 2.To find the optimal way to transplant the tissue to the rat heart with myocardial infarction.The different binding extracellular matrix protein will be evaluated for strong tissue attachment. 3. To assess the retention of transplanted tissue ring, to evaluate the improvement of the heart function after tissue ring transplantation.
