Au/silk NPs were obtained in narrow size distribution, with the surface charges can be modified in a similar method of modifying silk NPs. The obtained NPs show moderate drug loading, good stability in biological environment and negligible toxicity at cellular level. When irradiating the drug loaded NPs suspension at a wavelength similar to the absorption band of Au, it dramatically increased the drug release. More important, while drug release is relatively slower at pH7 in the case of silk only, we can obtain the drug release to almost 90% by increasing the irradiation time or increase the Au contents in Au/silk NPs. Under irradiation of a suitable laser wavelength, the photon energy at the SPR peak is absorbed effectively by Au NP and converts to thermal energy.

理由
Most targets of the research plan are nearly complete. The research is progressing to the investigation of in vitro behaviors of these nanocarriers with various types of drugs loaded. For Au/silk NPs, the pH- responsive drug release was associated with functions of silk side groups, while currently the photothermal-responsive drug release behavior is initially considered to be related to silk protein’s swollen and biodegradation. The possible synergistic effect between pharmaceutical activities of anticancer agents and the hyperthermia will be further confirmed in vivo.

今後の研究の推進方策 |
The research is currently focused on reproducing the results, especially for in vivo experiments, while building the next project to further develop the obtained knowledges to apply in 3D tumor model.

次年度使用額が生じた理由 |
To secure travel expenses for several international conference presentations which were postponed to 2022 due to COVID-19, part of the FY2021 budget was transferred to FY2021. In addition, another part of transferred budget is going to be for purchasing animal models for confirm the reproducibility of the research results in in vivo experiments.
