In this project, we aim to reconstruct a thin piece of skin equivalent with dermis and epidermis layer, which is easy to investigate under microscope, and is effective as a on-chip skin barrier.
Here, in the last fiscal year, the protocols for the formation of the skin-section chip was further optimized and the method for applying air-liquid interface culture of the epidermis on a microchip was established.
With the proposed device design and culture protocol, we were able to reconstruct a skin-section chip with differentiated epidermis, as confirmed both using phase contrast image and immunofluorescence imaging. In addition, electrical measurement(TEER) to evaluate the barrier function was also conducted.
With the support of this project, several journal articles, conference papers were published and a patent was filed.
