The BRAFV600E mutation is well known as the commonest and TERT-promoter (two hot spots, C228T and C250T) are known as a poor prognostic predictor genetic alteration found in adult PTCs, with geographic distinctions of its incidence and clinicopathologic significance. This study is the first report to reveal the association between genotype and phenotype incidence in adult PTC in the Japanese and Kazakh populations. Surgically resected PTCs from 342 Japanese and 211 Kazakh patients were analyzed for BRAFV600E and TERT-promoter mutations by droplet digital PCR (ddPCR) and Ki-67 LI was counted on IHC. The study demonstrated TERT-promoter (C228T and C250T) in 72 (21%), BRAFV600E 280 (82%) in Japanese PTC from Japanese population, and TERT-promoter (C228T and C250T) in 11 (5%), BRAFV600E138 (65%) in PTC among Kazakh population. Associations between molecular factors and Ki-67 LI, and clinicopathologic factors currently under the statistical evaluation. Publication of obtained results under preparation.
