The observation of kinesin using a high-speed atomic force microscope combined with total internal reflection fluorescence microscope was successfully conducted. It was possible to simultaneously observe the motion of kinesin on the scale of several tens of micrometers and gain detailed information of kinesin movement on the nanometer scale using the high-speed atomic force microscope. Further, the combined setup was improved so that prolonged observation on the scale of minutes was enabled. In the next steps, more data will be collected and experiments with microtubules containing defects will be conducted. The possibility to generate defects in a microtubule using a high-speed atomic force microscope has already been proven previously. It is planned to continue to collect data and publish the results in the next half year.
