Last year, I investigated the in vitro/in vivo effect of several novel materials on odontoblast-like cells (MDPC-23), human dental pulp stem cells (hDPSCs), and animal as planned. The in vivo study of CCN2 on wistar rats is in-progress. Mechanical pulp injury model was established on wistar rats. The histomorphological changes of dental pulp tissue of wistar rats at different time points after treated with CCN2 will be observed after collecting the tissue. I will evaluate the therapeutic effect of CCN2 on injured dental pulp tissue, and provide evidence for further clinical application. Besides, I checked some other novel materials on MDPC-23 and hDPSCs. Including autologous fibrinogen glue (AFG) and calcium salt of 4-methacryloxyethyl trimellitate (CMET). AFG enriched with growth factors is prepared from blood, it is easy to obtained using only simple centrifugation. I found that AFG promoted odontoblasts proliferation and adhesion when coated on the culture plate surface, high concentration of AFG also induced odontogenic/osteogenic differentiation of MDPC-23 cells. And the comparative study demonstrated that the inducing effect of CMET on odontoblast cells is much comparable to calcium hydroxide and MTA, which is routinely used in clinical. The results showed these materials exhibited strong promote effect on cell odontogenic differentiation. I will continue the futher investigation.

理由
The in vivo effect of CCN2 on injured dental pulp tissue will be comfirmed after collecting the samples I have operated on wistar rat molars. HE staining and immunohistochemistry staining will be performed after the sample is ready for collecting. On the other hand, I investigated the in vitro effect of some novel materials on MDPC-23 and hDPSCs, the result showed these materials are extremely promising for vital pulp therapy as it may induce dentin remineralization and pulp regeneration. Some of the data was complete written into paper and is under submission to SCI journal.
