研究概要

関節炎や関節破壊の病態形成に深く関わっているIL-1に焦点を絞り、IL-1およびその関連・下流遺伝子の免疫系、神経系、内分泌系における機能を明らかにし、生体システムの恒常性維持や疾患発症における役割を包括的な理解する。またこれらの解析を通じて生体内評価をした遺伝子改変マウスをバイオリソースとして広く研究者に供給することを目的とした。本プロジェクト期間中に新規5系統の遺伝子欠損マウスの作製に成功し、本研究開始時に作製済みであった4系統の遺伝子欠損マウスとあわせて戻し交配を完了した。これらの遺伝子欠損マウスの解析からIL-1関連遺伝子と関節炎などの自己免疫疾患や骨代謝制御との関連を明らかにし骨代謝制御異常のモデル動物を樹立した。またIL-1の中枢神経系における作用を解析し精神疾患病の新規モデル動物である可能性を示した。さらにIL-1が脂質代謝系の恒常性維持に関与していることを明らかにした。新規に作製したマウスを含めた遺伝子改変マウスの供給体制を整え、バイオリソースとして公開・配布を行い当該研究領域の発展に大きく貢献した。
