研究概要

相同組換えは、抗がん剤(シスプラチン、カンプトテシン)によって生じたDNA損傷を修復する機構の1つであり、50種類以上の分子が関与する。我々は、ニワトリDT40細胞で遺伝子破壊を実施することにより、以下のタンパク分子の、相同組換えにおける役割を解明した。解析した分子とは、RAD51補助因子(BRCA1, BRCA2, Palb-2, RAD51 paralogs (5種類), Rad52, Sfr1, Sws1)、DNA合成酵素、DNA切断酵素(CtIP, Fan1, RAP80, Slx4)である。
