光誘導タンパク質機能阻害法による染色体微細構造の顕微解析

研究概要

染色体微細構造において機能するタンパク質のin vivo機能解析をライブイメージングと光刺激の融合技術を開発した。特定の波長を持つ光を照射することにより活性酸素ROSを発生する光活性化タンパク質を用いて、微細構造特異的に染色体タンパク質を機能阻害し、その相互作用を時空間的に明らかにした。これにより、光誘導タンパク質機能阻害法を用いて、染色体形成初期の微細構造構築に寄与するタンパク質機能を同定することを示すことができた。
