研究概要

細胞内の蛋白質の立体構造形成を助ける分子シャペロンの一つである、大腸菌シャペロニンGroELは、ATPと結合することによってその機能を発揮する。本研究ではGroELへのATP結合を光親和性標識、アミノ酸配列分析、ストップトフロー蛍光スペクトル、滴定型熱量計などを用いて調べた。その結果、(1)Tyr360近傍に2つ目のATP結合部位があること、(2)カリウムイオン非存在下では第1結合部位へのATP結合のみが観測され、その結合定数は10^4M^<-1>であること、などが明らかとなった。
