好塩基球によるTh2誘導機構とアレルギー慢性化の解析

研究概要

好塩基球は末梢白血球中1%以下存在する少数派細胞で、FcεR1を構成的に発現し、アレルゲン-IgE複合体で活性化されると、直ちにヒスタミン、脂質メデイエーター、IL-4、IL-13などを産生して、即時型過敏症を惹起する。私達は本細胞がIL-18とIL-33受容体を発現し、これらのサイトカインで刺激されるとIL-4、IL-13などを産生することを明らかにした。また、好塩基球がMHC class IIとCD80/86を発現し、抗原蛋白を取り込み細胞内でプロセス後にMHC class IIに抗原ペプチドを結合させ、同時にIL-4を産生することで、ナイーブTh細胞をTh2細胞に分化誘導することを明らかにした。
