研究概要

本邦では欧米に比較して肝炎ウイルスの汚染率が高く,急性肝炎あるいは肝線維症(慢性肝炎・肝硬変)から肝不全で死亡する例が多く,大きな社会的問題となっている.肝移植が頻繁に施行されるようになった現在でも,提供肝の絶対数は不足しており,多くの肝不全患者を救うためには新たな治療法の開発が必要である.我々は,肝不全患者の治療戦略として,(1)抗肝線維化療法,(2)肝再生療法,(3)分裂寿命/細胞死からみた肝細胞庇護療法を3つの重要な開発研究課題と位置づけ,集学的な研究を遂行してきた.肝の線維化には,類同壁細胞である肝星細胞(hepatic stellate cell : HSC)が重要な役割を担っている.当該研究課題では,HSCの形質転換時にクロマチン修飾の変動する転写領域を網羅的に同定し,個々の遺伝子発現がHSCの表現型にどのような影響を与えるかギガベース(10億ベース)シークエンサーを用いた,ChIP (chromatin immunoprecipitation) sequencing法によって解析した.活性型HSCの分離を行い,形質転換実験をHDAC(histone deacetylase)阻害薬,interferon (IFN)-γを用いて基礎実験を行った.いずれでも,形質転換が可能であり各々の条件でのDNA, mRNA,蛋白質の解析を行った.形質転換時のエピゲノム形成に重要な,多能性維持関連蛋白質の特定に至った.多能性維持関連蛋白質のうちnucleus accumbens associated-1はエピゲノムの保持に重要な蛋白質であり,MYC, Nanogとの相互作用があることが明らかとなった.
