血清中ヘパラナーゼ測定法の確立と抗ヘパラナーゼ薬による癌転移抑制療法の開発

研究概要

血清中Heparanase値測定法の確立のため、ELISA 反応確認試験として、抗HPR1抗体と合成ペプチド、ビオチン標識ペプチオドを用いたELISAを構築し、抗ウサギ抗体を固相したマイクロプレートを用い、抗体と各ペプチドの反応が確認できた。HPR1ペプチドとして10～1,000nmol/lの範囲で濃度に依存する検量線を得ることができた。HPR1 ELISAキットを用いて血清サンプルの測定を行った結果、良好な添加回収率、希釈直線性が得られた。本キットを用いて血清中のHPR1を定量することが可能であると判断された。さらにELISA 性能評価試験では、HPR1ペプチドに対する抗体と合成ペプチドを用いたELISAを検討した結果、良好な反応性を確認できた。HPR1ペプチド濃度として1～100nmol/lの範囲で定量が可能であった。血漿を用いたELISAも良好な反応を示し、血液サンプル測定への可能性が示され、HPR1タンパクあるいは実際のサンプルを用いた検討を行うことで有用性の検証を行うことが可能となった。HPR1 ELISA キットを試作し血清サンプルの測定を行った結果、検量線の範囲に入る良好な結果が得られ、臨床応用が可能と判断された。
