研究概要

制御性T細胞の分化・機能制御メカニズムを明らかにすることを目的として二つの研究課題に取り組んだ。第一に、ヒト自己免疫疾患IPEXにおいて同定されているFoxp3遺伝子変異を導入したノックインマウスを作製して変異の制御性T細胞分化・機能に与える影響を解析した。一つの変異マウスでは、自己免疫疾患の発症と相関して末梢組織に局在する制御性T細胞サブセットが選択的に欠損することを見出し、この制御性T細胞サブセットが自己免疫寛容の成立・維持に重要な役割を担っていると考えられた。第二に、正常個体に存在するFoxp3^+ T細胞は不均一な集団であり、一部は環境からの擾乱に対してFoxp3発現を失ってヘルパーT細胞へ分化する可塑性を示すことを明らかにした。
