17型コラーゲンの分子イメージングによる水疱性類天疱瘡の病態メカニズムの解明

研究概要

本研究により、水疱性類天疱瘡(BP)患者IgG自己抗体が表皮基底細胞膜上に発現している17型コラーゲン(COL17)へ結合したのち、ダイナミックに細胞内へ取りこまれることが明らかとなった。COL17の細胞外領域は細胞膜近傍で切断されるが、BPの自己抗体が、切断された後に生じる新規抗原エピトープに対し反応するものがあることも解明した。更に、COL17は、細胞外マトリクス中のラミニン332やIV型コラーゲンと相互作用しており、表皮細胞はCOL17を切断しながら遊走することも明らかにした。
