研究概要

申請者は、DNA中メチル化シトシンの簡便検出法の開発を目的として、光反応を用いたメチル化シトシン選択的な鎖切断システムの構築を試みている。これまでに、ナフトキノン誘導体を光増感剤として使用すると、DNA中のメチル化シトシン部が酸化されることを見出し、当該部分で選択的な鎖切断が起こることを明らかにしている。
本研究において、反応の詳細を調べたところ、メチル化シトシン塩基の光酸化に伴い、ホルミルシトシン誘導体が生成していることがわかった。続いて、反応メカニズムを考察したところ、一電子酸化反応に続く脱プロトン化とメチルラジカル生成が本酸化反応には関与していることが示唆された。
