椎間板内在性幹細胞の解析と変性への関与、治療への応用へ向けた基礎的研究

研究概要

大小動物およびヒト検体を用いた椎間板細胞を検討した結果、髄核細胞の"heterogeneity"とその特色を明らかにした。また椎間板内における組織内在性幹細胞とそのニッチにつき解析、椎間板内在性幹細胞やその未分化度、活性度について年齢と負の相関関係にあることを実証した。これは椎間板再生により適した細胞集団の選択に役立つものと考える。さらに他臓器では骨髄などの幹細胞プールより細胞が"mobilize"してくるが、椎間板においてそのような機構が存在するかにつき、GFPキメラマウスを用いて解析した。その結果、骨髄から"mobilize"する細胞は終板付近には多く認められるが、線維輪、髄核と中心部に行くに従い減少し、椎間板には幹細胞プールからの細胞供給が極端に少なく、このことは外来から細胞を補充することの妥当性を示した。
