研究実績の概要 |
This project aims the identification of microbes associated with sucking type rice plant pest brown planthopper (BPH). Previously, we established that these microbes can elicit rice defense against BPH. In the current project, we are collecting BPH from various areas in Japan and explore properties of their microbe-containing honeydews. We are also screening for best biologically active honeydew samples in rice, and isolate bioactive microbes from these honeydews to establish methods for microbes to be used as novel tools for biological protection of rice against herbivores.

The experiments progress as expected. We completed main tasks planned for initial 6 months of the project. There is no problem to report at present.

今後の研究の推進方策 |
In future work, we will continue isolating microbes from stored honeydew samples and keep subjecting them to sequence analysis for identification purposes. We will continue testing the activity of microbes on rice immune defense systems, using cultured rice cells and intact rice seedlings. Furthermore, isolated DNA from honeydew samples will be subjected to Next Generation Sequencing (NGS) for gaining the information about overall diversity of rice brown planthopper honeydew-associated microbiome. In summer months in 2021-22, two field trials will be conducted by applying selected microbial strains isolated from brown planthopper honeydew to rice plants to gain information about rice defense and resistance level under natural conditions. Screening for insect density and damage will be conducted weekly. In parallel, glasshouse experiments will be conducted under semi-controlled cultivation conditions to precisely asses the effect of microbes on plant defense, pest survival and reproduction. The results will be summarized and presented in scientific meetings in 2022. Summarizing of all results and publication of results is expected to be completed in 2023.
