研究実績の概要 |
To achieve an effective enzyme combination for enzymatic hydrolysis of laminarin and alginate in kelp, a glycoside hydrolase family 55 enzyme (SactELam55A) and two polysaccharide enzyme families (PLs), seven PL17 and two PL18 were successfully prepared using a recombinant enzyme production method. The reaction optima of temperature and pH of these enzymes were individually determined for combinatorial enzyme hydrolysis. Additionally, one reported PL7 enzyme was gene synthesized to produce monomer from alginate polymer in combination with PL17 and PL18 alginate depolymerizing enzymes.

理由
Four classes of kelp depolymerizing enzymes have been produced and determined their reaction optima and ready for kelp hydrolysis. Also, four different kelp species have been collected and fractionated for enzyme-assisted kelp hydrolysis. We are a bit behind in terms of publishing the first paper for kelp hydrolysis using a combination of two enzymes, SactELam55A and PL18.

今後の研究の推進方策 |
All enzymes will be subjected for combinatorial kelp hydrolysis to determine the most efficient enzyme combination under reaction optima (Temperature and pH). Kelp will be hydrolyzed by the optimum enzymatic hydrolysis, and hydrolysates will be analyzed by analytical methods using HPLC and GC/MS. We will also fractionate hydrolysate to determine end products. Fractionated samples will be tested for their health beneficial functions.
