Extracellular vesicles (EVs) are classified into three subclasses, including exosomes, microvesicles (MVs), and apoptotic bodies. Currently, there are no methods to distinguish exosomes from samples containing different EVs. We have developed a method to distinguish exosomes from other extracellular vesicles based on their changed states and mechanical properties, which is crucial for separating exosomes from a heterogeneous sample. We have identified exosomes from heterogeneous EVs using both an anion exchange column based on their surface charge and AFM nanomechanical measurements based on their Young's modulus differences. From the nanomechanical measurements, we found that exosomes have higher Young's moduli than MVs.
