研究実績の概要

The role of GPR61 in the regulation of Adiponectin expression was confirmed in the intestinal cells by establishing general GPR61 knockout mice as the knockout mice had less adiponectin in the intestinal cells compared to the wildtype mice. Plasmalogens (Pls)-mediated increase of Adiponectin expression in the intestinal epithelial cells of adult mice has been confirmed by primary culture of the intestinal cells. Oral ingestion of plasmalogens reduced the fatty liver in the ZDF rats associated with the increase of Adiponectin in blood serum. We also checked various gene expression related to fatty acid biosynthesis and metabolism in the liver tissue of ZDF rat and found some candidate genes of plasmalogens, which could be the mechanism of Pls-mediated reduction of the fatty liver.

理由

Some part of the projects are progressed very well and I could also successfully make general GPR61 knockout mice to support my research findings. However, I am delayed to make Intestinal tissue-specific knockout mice due to some problems in the materials.

今後の研究の推進方策

By using the general knockout mice of GPR61, I will study the role of plasmalogens in the regulation of Adiponectin and fatty acid metabolism in the liver. I will study the detail lipid biosynthesis pathway to find out the mechanism of how plasmalogens can reduce fatty liver. I will also try to make the intestinal tissue-specific GPR61 knockout mice.
