研究実績の概要 |
This year, we sampled scallop in Akkeshi, Japan. We collected 150 individuals while dredging and fixed samples in methalcarn for histological work, and preserved some of the tissue and organs from the individuals for DNA extraction. All samples were photographed and measured. In the laboratory, genomic DNA was extracted and stored for future genomic work. Samples were sectioned and stained with Hemotoxylin and Eosin. Three main parasitic infections were identified: ciliates, bacteria, and Apicomplexans (probable Nemotopsis). Samples were further processed with a Laser dissection microscope and genomic DNA was extracted. PCR were performed to give a molecular identity to the infections. Currently, we are comparing these histological sections and PCR-generated sequences to genomic data collected during a previous project in 2017. Our goal is to summarize these findings and submit our results for publication in an international journal.

理由
The covid-19 infections delayed some of our field work, and limited our ability to use the marine laboratory in Akkeshi. During the Summer of 2020-2021, Hokkaido University had a raised biosafety status which limited the ability of people to work in the laboratory together. Therefore, we had trouble conducting some fieldwork, and conducting work together in the lab at the university.

今後の研究の推進方策 |
In August 2022 and December 2022, we will visit Akkeshi Marine Laboratory. Our aim to to collect 150-200 more scallop and confirm the pathology, specifically looking for ciliate, bacteria and apicomplexan infections. Similarly, we would like to conduct more detailed morphological identification of the parasitic organisms. Our initial findings suggest that these are likely new species, and so our goal is to place these into a phylogenetic context and also to provide a scientific name for these new species. Moreover, we are interested in conducting a more detailed genomic survey of the infections. To this end, we will examine infection rates, and seasonality of these infections.

次年度使用額が生じた理由 |
Due to Covid-19, we have moved sampling trips from August and December 2021 to August and December 2022, respectively. The funding will be used to pay for travel, collection, and the processing of samples during these sampling trips, and for processing samples at the laboratory.
