In oder to elucidate the function of Nod2 and dysbiosis in the ileum we set out the followings during the first year:
1) establishing in vitro analysis of Paneth cell function for bactericidal activity. We established the in vitro system to isolate Paneth cells, which is ready to secrete alpha defensins upon ligand stimulation. The molecular mechanisms by which Nod2 regulate the section will be sought out at the second year.
2) establishing real time secretion assay of Paneth cells. With Dr. Ayabe's group collaboration, we set up real-time imaging system which provides robust information as to alpha defensin secretion through real-time visualization. The system will be applied to the Paneth cells derived from wild-type, Nod2- or Rip2-deficient mice at the second year.
