Previous study, I found that in-house mice have different microflora profiling. Therefore, I could not compare microflora profiling after stress induction. In this time, I bred mice from only 2 families. Then, I kept all mice in 1 cage for 2 weeks. Then, 12 mice were separated into each new cage for 2 weeks (control=6 mice, stress=6 mice). Then, mice were induced stress for 6 weeks. Feces were collected every week. Plasma was collected after 6 weeks of stress induction. The rhythm clocks were clearly different. The stress hormones were determined from plasma. The cortisol level of stress mice was significantly increased. While, Adrenaline and noradrenaline were not significantly changed. I also analyzed metabolome in plasma and feces. However, I could not find any significant differentiation. Microflora analysis was determined from feces of 0-, 1-, 4- and 6-weeks stress. I could not see the group of microflorae by PCOA ordination. Whereas, I found that abundant genera of Enterococcus, Lactoccus and Unas Prevotellaceae were significantly increased in stress mice at 1 ,4 and 6 weeks. Steptococos was significantly increased in stress mice at 1 week stress, and then decreased at 4 and 6 weeks. Unas Desulfovibrionaceae was increased in stress mice at 4 and 6 weeks.

I can determine biochemistry and function of stress mice as plan.

今後の研究の推進方策

Although, I kept mice together for 2 weeks before experiment. Whereas, I could not see the group of microflorae by PCOA ordination since 0 week. My previous study showed that all mice from one breeding company has very similar microflora profiling. Therefore, I will use mice from this company to do stress model again. I will also determine cytokines profiling from brain, spleen, mesenteric lymph nodes, peyer's patch and liver.

次年度使用額が生じた理由

I used AIST before I could get kakenhi. I need to do analysis of metabolome and microflora again that need much expense more than a plan. Therefore I will use this year grant to do analysis of metabolome and microflora again. I will buy machine for 2022 year`s experiment.
