Collagen microfibers size effect on the reconstruction of vascularized adipose tissues for soft tissues regeneration applications

研究実績の概要

The first aim of this research was to investigate the use of collagen microfibers to reconstruct fat tissue. The right microfibers characteristics were found to get adipocytes maintenance and blood vessels construction in the ball tissues. Next, these balls could be injected and merge together with vasculature interconnection to form bigger tissues. Following their implantation in mice, they showed a higher cell survival and volume maintenance after 3 months, compared to lipoaspirated fat and non-vascularized balls injections. This was explained by their greater amount of blood vessels which connect to the host blood vessels, also involving lymphatic and neural networks. Finally, the cryopreservation possibility was confirmed, enabling later reinjection for controlling the graft volume.

理由

These results were obtained using purchased endothelial cells, which is easier, but these cells came from another patient. It enabled us to progress quickly up to the step of the mouse implantation. These results are important to prepare the future plans where it will be only necessary to use different cells. Actually, in order to fulfill the requirement of only autologous cells implanted as planned in the research purpose, we did tried the isolation of the endothelial cells from the same patient tissue. But the process appeared difficult for getting a high number of these cells, enabling their use in the model. So, the differentiation into endothelial cells using the adipose stem cells was also investigated and gave promising results which will be used for the next step.

今後の研究の推進方策

The future work will focus on the use of autologous cells only for the construction of the adipose ball tissues by improving the adipose endothelial cells isolation or the endothelial differentiation from the adipose-derived stem cells, as already tried.
Then, the next focus will be on the scale-up of the current process which was performed manually, by using a bioprinter for the seeding of the adipose ball tissues and then culture them in a bioreactor in high number together.
In parallel, three publications were written on the adipose tissues construction, one was published (doi:10.34133/2021/1412542) and the two other are still under revision and should be accepted in the next fiscal year.
With the results of the full autologous model, another manuscript will be considered.

次年度使用額が生じた理由

The main reason for the remaining of money for next year is due to the fact that the international conferences were not possible during the fiscal year 2020, due to the global Covid-19 pandemic, leading to a much lesser use of the travel fees compared to that was planned. Instead of travelling we did only online international conferences or free national conferences.
