フラボノイドの高機能化に関わるプレニル化酵素遺伝子ファミリーの機能解剖と酵素工学

研究概要

フラボノイド特異的なプレニル基転移酵素SfN8DT-1の配列を元に、さらにクララ培養細胞からイソフラボンに特異的なSfG6DT、カルコンに特異的なSfiLDTを取得した。これらはいずれも、9回の膜貫通ドメインとプラスチド配向性シグナルを持ち、ビタミンEの生合成酵素ファミリーに類似であることが判明した。またダイズのファイトアレキシン生産に重要なプレニル化酵素GmG4DT、またマメ科以外のオオバギ(トウダイグサ科)からもそれぞれ基質ならびに生産物特異性の高い酵素をクローニングした。さらに性質の異なるSfN8DT-とSfG6DTの間でキメラ酵素を作成し、基質認識部位の絞り込みに成功した。またプレニル化化合物の代謝工学を、植物および微生物ホストにおいて行った。
