研究概要

RNAi(RNA干渉)は、遺伝子機能阻害のための頻繁に使われる技術である。この現象が初めて線虫C. elegansで発見された当初より、二本鎖RNAが細胞間を伝播することが知られており、SID-1という分子は、伝播するRNA干渉の際の二本鎖RNAの取り込みに必要であるとされる少数の蛋白質をコードする。我々は、エンドサイトーシスメカニズムが取り込みに関わっている可能性を検討するために、vps-45遺伝子変異体を調べたところ、ほとんどの遺伝子については弱い表現型であったが、gon-1遺伝子RNAiを行うと、抵抗性であった。GON-1蛋白質はメタロプロテアーゼをコードし、DTCと体壁筋での蛋白質の分泌に必要な分子であることが判明した。また、この分泌機能には、GONドメインのみが必要であることが明らかになった。
