研究概要

リコンビナントSULT2A4を大腸菌で発現し、基質特異性を検討した。その結果、胆汁酸類のC-7位の水酸基を特異的に硫酸化することが明らかとなった。次に、反応機構解明のため、触媒中心に位置するLeu99およびHis48を標的として部位特異的変異の導入を行い、変異酵素を作製し解析を行った。その結果、全ての変異酵素でVmax/Kmの値が低下した。さらに、L99H、H48T、H48NはKm値を著しく大きくしたことから、基質との親和性に関わる残基であると判明した。
