研究概要

軟骨内骨化は骨格成長や変形性関節症(OA)発症に関与する。今回、転写因子CCAAT/enhancer-binding protein(C/EBP)ファミリーの生理的および病的な軟骨内骨化における役割とについて検討した。
C/EBPδホモ欠損マウスおよびC/EBPε欠損マウスにおいては明らかな骨格形成異常は認められなかったが、C/EBPβホモ欠損(-/-)マウスは同胞野生型(+/+)マウスに比べて胎生期より成長障害が見られ、肢芽でのCOL10、MMP13の発現が低下していた。またC/EBPβヘテロ欠損(+/-)マウス(8週齢)の膝関節に実験的OAモデルを作成した場合でも、+/+マウスに比べて関節軟骨でのCOL10、MMP13の発現の低下と共に軟骨変性が抑制された。C/EBPβはRunx2のco-factorとして知られているため、C/EBPβとRunx2の複合遺伝子欠損マウス(C/EBPβ-/-; Runx2+/-)を作成したところ、同胞C/EBPβ-/-マウスよりも強い成長障害を認めた。組織学的検討には、軟骨内骨化の遅延および一次海綿骨量の低下を認め(alcian blue/von Kossa二重染色)、COL10の発現はC/EBPβ-/-と同程度であったがMMP13の発現が著明に低下していた。このメカニズム解明のため、ヒト軟骨系細胞株SW1353にC/EBPβとRunx2を共導入すると、増殖能(CCK-8 assay)は単独導入と同程度であったが、各種軟骨分化マーカーの中でMMP13の発現(real-time RT-PCR)が相乗的に上昇した。一方、C/EBPβとRunx2の発現は相互の強制発現によって変化はなく、独立した制御が示された。MMP13 promoter-luciferase gene construct導入SW1353にC/EBPβとRunx2を過剰発現させたところ、協調して転写活性を促進することが示され、更に段階的deletion、mutagenesis、tandem-repeat constructを用いた検討によって、MMP13遺伝子の転写開始点上流-144～-89 bpの部位にOSE2に加えC/EBPモチーフを含む応答領域を同定した。C/EBPβはRunx2と協調してMMP13を転写誘導し、骨格成長やOAの軟骨変性を制御していることが示された。C/EBPβ、Runx2の相互作用は成長障害およびOAなどの軟骨変性疾患などの治療ターゲットとなりうることが示唆された。
