研究実績の概要 |
The aim of this project is to shed light on Blattabacterium genome evolution. To do so, we planned to sequence a series of new Blattabacterium genomes. During FY2021, we sequenced fat body DNA from about 300 cockroaches using NovaSeq. We assembled the 300 libraries and obtained about 100 full Blattabacterium genomes. The remaining 200 genomes were fragmented and need to be resequenced. This will be done during FY2022. The full Blattabacterium genomes we obtained have been annotated and are ready to use for comparative genomics analyses.
We published one paper on Blattabacterium during FY2022. The paper was published in Molecular Biology and Evolution (IF: 16.2). The reference of the paper is as follow: Kinjo Y.*, Lo N.* (*equal first authors), Villa-Martin P., Tokuda G., Pigolotti S. & Bourguignon T. (2021) Enhanced mutation rate, relaxed selection, and the ‘domino effect’ are associated with gene loss in Blattabacterium, a cockroach endosymbiont. Molecular Biology and Evolution, 38, 3820-3831.

理由
In my project, I planned to sequence 250 Blattabacterium genomes during FY2021-2023. We managed to sequence about 100 full Blattabacterium genomes during FY2021, which was as expected. I also planned to start the data analysis, which we did. All sequenced libraries have been assembled and the complete genomes of Blattabacterium have been annotated. Although I did not originally plan to publish any paper during FY2021, we did publish one paper in a high-impact journal. In this paper, we reanalyzed some genomes we previously published from a new angle. Therefore, the genomes we have produced so far in this project have not been published yet. They will be published during FY2022 and FY2023.

今後の研究の推進方策 |
During FY2022, we will continue sequencing additional genomes of Blattabacterium. These genomes will allow us to explore the relationship between gene loss, dN, dS, dN/dS, and GC-content. I expect to have sequenced at least 250 full Blattabacterium genomes during FY2022. Although, we already started determining the relationship between gene loss, dN, dS, dN/dS, and GC-content, we will explore these relationships in more details during FY2022. I expect we will have carried out most of the data analyses and will be in the process of writing scientific paper by the end of FY2022. The papers will be published during FY2023.
