Our research group has developed a micro drawing device, the chemical pen, which enables to control solution in local area on surfaces at the nanoscale using confined open-space laminar flow. We have developed an open-space laminar flow approach for fabricating a single gold nanowire at precise position under normal condition. This method enabled us to fabricate gold nanowires with the widths ranged of 200 nm to several ten micrometers and lengths ranged of 3 mm or less at targeted locations. The gold nanowires were composed of multiple nanoparticles aggregated into a wire-like structure. The fabricated gold nanowire was demonstrated an excellent biosensing for IgA with an extremely low limit of detection (1 aM). Thus, this novel technique is expected to be applied in a wide range of fields. It is the first time to fabricate gold nanowires at precise position and in normal environment. Compare to Ag nanowire, gold nanowire is more widely used for biosensing. The high-performance IgA detection was achieved using a single gold nanowire in this study, however, by changing the modification of the gold nanowires according to the target substance, we expect to develop a sensor that can measure various substances with high sensitivity in the field of pharmacology, medical care, food analysis, environmental analysis and even of infection diagnose.

今後の研究の推進方策 |
The setup of resistance recording system. The detection system involves a conductive chamber, a nanowire sensor, an I/V converter, A/D converter and a computer for record. A preparing nanowire sensor and A/D transformer will be set in a conductive chamber for eliminating electromagnetic interference. Detection of CEA from a single cell using the nanowire sensor. Caco-2 cell are cultured in petridish with low cell density for use. Before detection of CEA, the culture medium in the petridish will be replaced with fresh culture medium. The nanowire sensor that have connected with resistance recording system will be manipulated close to the target cell. Then, the resistance change will be recorded by software in the computer.
