This research develops the first example of hydroxyl-radical-responsive protein labeling reagents for fluorescence imaging and proteomics, which aims at the comprehensive understanding of hydroxyl radical in living systems by unraveling its localization and surrounding/associated proteins. In the first year, a series of phenol/aniline-derived chemical probes have been designed and synthesized and in vitro protein labeling experiments successfully identified hydroxyl-radical-selective probes. In the second year, we moved to live-cell assays with synthesized cell-permeable probes. The probe structure was slightly modified to reduce the background labeling in living cells. Preliminary results have demonstrated its availability in ROS conditional protein labeling in activated macrophages.
